T4 gene 32 protein

T4 Gene 32 Protein is a single-stranded DNA-binding protein required for DNA replication and repair in T4 Phage. It coordinately binds and stabilizes the ssDNA region that transiently forms, and plays an important structural role in the T4 Phage DNA replication. The protein has also been widely used to stabilize and label ssDNA regions in order to observe intracellular DNA structure with an electron microscope. The latest reports have shown that T4 Gene 32 Protein can promote digestion of restriction enzymes, improve the efficiency of reverse transcription in RT-PCR, enhance the activity of T4 DNA polymerase and increase the yield of PCR.

Storage Temperature

Store at -20 ℃, valid for one year (Avoid repeated freeze-thaw cycles).

Storage Buffer

300 mM NaCl, 10 mM Tris-HCl (pH 7.5 @25°C), 1 mM DTT, 0.1 mM EDTA and 50% glycerol.

Protein Concentration

The concentration of T4 Gene 32 Protein is 10 mg/mL±5% as determined by UV absorption at 280 nm.

Molecular Weight of Protein

The molecular weight of T4 Gene 32 Protein is 33.5 kDa.

Heat Inactivation

65℃ for 20 minutes.

Quality Control

Exonuclease Activity: 10 μg of T4 Gene 32 Protein with 1 μg λ-Hind III digest DNA at 37℃ for 16 hours yields no degradation as determined by agarose gel electrophoresis.

Endonuclease Activity: 10 μg of T4 Gene 32 Protein with 1 μg λDNA at 37℃ for 16 hours yields no degradation as determined by agarose gel electrophoresis.

Nicking Activity: 10 μg of T4 Gene 32 Protein with 1 μg pBR322 at 37℃ for 16 hours yields no degradation as determined by agarose gel electrophoresis.

RNase Activity: 10 μg of T4 Gene 32 Protein with 1.6 μg MS2 RNA for 4 hours at 37℃ yields no degradation as determined by agarose gel electrophoresis.

E. coli DNA: 10 μg of T4 Gene 32 Protein is screened for the presence of E. coli genomic DNA using TaqMan qPCR with primers specific for the E. coli 16S rRNA locus. The E. coli genomic DNA contamination is ≤1 E. coli genome.
